Chew gum, lose weight? Hormone that helps people feel 'full' after eating can be delivered into bloodstream orally

ScienceDaily (Nov. 21, 2011) ? Most people understand that serious weight loss requires changing attitudes toward what they eat and how often they exercise. But, what if the process could be aided by simply chewing a stick of gum after meals? That's the question a team of scientists, led by Syracuse University chemist Robert Doyle, is trying to answer. In a groundbreaking new study, Doyle's team demonstrated, for the first time, that a critical hormone that helps people feel "full" after eating can be delivered into the bloodstream orally.

Doyle's study was published online Nov. 4, 2011 in the American Chemical Society's Journal of Medicinal Chemistry and is forthcoming in print. Doyle is an associate professor in the Department of Chemistry in SU's College of Arts and Sciences. He collaborated on the study with researchers from Murdoch University in Australia.

The hormone, called human PYY, is part of a chemical system that regulates appetite and energy. When people eat or exercise, PYY is released into the bloodstream. The amount of PYY that is released increases with the number of calories that are consumed. Past studies have shown that people who are obese have lower concentrations of PYY in their bloodstream both when fasting and after eating than their non-obese counterparts. Additionally, intravenous infusion of PYY into a volunteer group of obese and non-obese individuals increased the serum levels of the hormone and lowered the number of calories both groups consumed.

"PYY is an appetite-suppressing hormone," Doyle says. "But, when taken orally, the hormone is destroyed in the stomach and that which isn't destroyed has difficulty crossing into the bloodstream through the intestines."

What's needed is a way to disguise the PYY so that it can travel through the digestive system relatively unharmed. Several years ago, Doyle developed a way to use vitamin B12 as a vehicle for the oral delivery of the hormone insulin. B12 is able to pass through the digestive system with relative ease and carry with it insulin, or other substances, into the bloodstream. Similarly, his research team attached the PYY hormone to his patent-pending vitamin B12 system. "Phase one of this study was to show that we could deliver a clinically relevant amount of PYY into the bloodstream," Doyle says. "We did that, and we are very excited by the results."

The next step involves finding ways to insert the B12-PYY system into such things as chewing gum or an oral tablet to create a nutritional supplement to assist individuals in losing weight in much the same way as nicotine-laced gum is used to help people stop smoking. "If we are successful, PYY-laced gum would be a natural way to help people lose weight," he says. "They could eat a balanced meal, then chew a stick of gum. The PYY supplement would begin to kick in about three to four hours later, decreasing their appetite as they approach their next meal."

Proposal to tax Jack Daniel's whiskey derailed (AP)

NASHVILLE, Tenn. ? Jack Daniel's officials are toasting the defeat of a proposal to tax whiskey at its celebrated Tennessee distillery.

The Moore County Council in Lynchburg, Tenn., voted 10-5 Monday evening to kill a proposal that could have taxed Jack Daniel's up to $5 million annually, with all the revenue going to local coffers.

"We hope we've been able to demonstrate that the distillery pays more than its fair share of taxes and that we've contributed to our way of life in Lynchburg," said Tom Beam, senior vice president and general manager of production at the facility.

The vote reversed an earlier one that had asked the Tennessee legislature to authorize a local referendum on the per-barrel tax proposal.

"We've educated the community a little more," Beam said Tuesday in a telephone interview from the distillery, located in the hills of south-central Tennessee. "They realized after we got our side of the story out how much we do."

The 145-year-old distillery and its employees, along with Lynchburg, have been the focus of Jack Daniel's folksy advertising for years. Bottles of the charcoal mellowed sippin' whiskey list Lynchburg's population as 361, but the town and county really have about 6,400.

The distillery, owned by Louisville, Ky.-based Brown-Forman, now pays $1.5 million in local property taxes.

"We hated to see this drive a wedge through our family here," Beam said. "This is our home and we'll try to do the right thing."

The Jack Daniel Distillery, with about 450 employees, is the largest employer in Moore County. The local Chamber of Commerce came out against the proposal at the meeting.

Supporters of the proposal said the issue is dead for now and they may quit trying.

"That's democracy in action, I suppose," Charles Rogers said of the vote after spearheading the proposal.

"I may bow out of this," he added. "But I still think people ought to have the right to vote on it (in a referendum)."

Ten million cases of the sour mash whiskey, led by Old No. 7, are sold worldwide every year, making it the No. 1 brand in sales globally.

"Our friends and neighbors around Moore County, the state, the country and even globally have been supportive," Beam said.

Company spokesmen never said whether the tax would have meant higher prices at the retail level.

Ironically, Moore County is dry, meaning Jack Daniel's cannot be sold legally in the county.

Really sharp looking, very original and an excellent premise! I'd love to submit the 'marksman' type character as I think I could do them justice, but I'm on my way to school at the moment, so profile creation is a bit beyond me. If you don't do reservations, then I'll just hold out for whoever is left. Maybe a former NPC? I don't know. I'd love to be a part of this.

Peter Thiel Stutters, Wants to Live Forever (The Atlantic Wire)

Earlier this year, we learned that early Facebook investor Peter Thiel didn't believe in higher education, and thanks to a just-published New Yorker profile, we now know that he also doesn't believe in death. What a contrarian! For the straight and narrow Americans out there, it gets even grittier. Thiel, pictured above with TechCrunch founder Michael Arrington, also invested in the development of anarchic off-shore colonies ? "sea-steading" ? through a nonprofit slash Randian fantasy founded by Milton Friedman's grandson. He wrote a big check for the Singularity Institute which, in New Yorker writer George Packer's words, "is preparing for the moment when a machine can make a smarter version of itself, and aims to insure that this 'intelligence explosion' remains 'human-friendly.'" Over the course of ten pages, including one full-page, full-color portrait of Thiel sitting on what appears to be a massive Ottoman made from the fur of exotic animals, Packer describes Thiel as nothing less than a genius and nothing more than a ginger-haired math nerd trapped in a billionaire's lifestyle. This is probably best communicated through Packer's slight but unavoidable focus on Thiel's voice, especially his stutter. Packer writes:

His most striking feature is his voice: something metallic seems to be?caught in his throat, deepening and?flattening the timbre into an authorita- tive drone.?During intense moments of cerebration, he can get stuck on a thought and fall silent, or else stutter for a full forty seconds: "I would say it's?it's? um?you know, it is?yes, I sort of agree?I sort of?I sort of agree with all this. I don't?um?I don't?um?there is a sense in which it's an unambitious perspective on politics." Thiel expresses no ill will toward anyone, never stoops to gossip, and seldom cracks a joke or acknowledges that one has been made.?

One can't help but suspect Packer is cracking a quiet joke about Thiel the wannabe robot whose actually a noticeably imperfect human. The bulk of the profile, however, focuses on the wannabe robot element of Thiel's personality and ambitions. We hear little about Thiel's background as a?hedge funder, but we learn a lot about his quirky branching out as a venture capitalist. The quest for immortality can be best summed up with one quote. Thiel told Packer, "Probably the most extreme form of inequality is between people who are alive and people who are dead."

Woman says her son a mentor to accused Ohio teen (AP)

COLUMBUS, Ohio ? The mother of a jailed Ohio man said Saturday her son has "a very caring heart" and was a mentor to a teenager charged in a deadly robbery scheme in which police say victims were lured with bogus help-wanted ads on Craigslist.

Carol Beasley told The Associated Press that she spoke to her son Richard and she prays that a newspaper report that he is a suspect is not true.

"The most I can say is this is just a big shock to us," she said in a telephone interview. "I pray it's some other person and not him."

Richard Beasley, 52, was in jail on unrelated charges when police arrested two suspects in the robbery scheme, including a 16-year-old boy Beasley's mother says he befriended. The teenager was charged Friday with attempted murder. A judge issued a gag order in the case, and the names of the two victims and the adult suspect, who so far has not been charged, were not released.

Authorities say a South Carolina man who responded to a bogus ad for a farmhand in Ohio was taken Nov. 6 to a desolate area and shot in the arm. He escaped and sought help at a farmhouse in Caldwell. The Columbus Dispatch identified the man as Scott W. Davis, 48, citing a Noble County Sheriff's Office incident report. Davis reportedly lived with his mother in Canton.

Last week, authorities found one hand-dug grave they believe was intended for the man who escaped and a second grave that held the body of a Florida man. The newspaper reported that paperwork in the case against the teenager says he was assisting a Richard J. Beasley in attempting to murder the man who survived.

Carol Beasley, 70, acknowledged that her son has a history of trouble with the law, but she was at a loss to explain where things went wrong. She said her son took the teenager to church almost weekly, took him fishing and to the movies or to play video games, and the two also did volunteer work delivering food to needy people.

"He worked as a street chaplain, did a lot of things like that, that's why all of this is difficult, you know?" the mother said. "I don't know, maybe sometimes when you work with street people, maybe you are just too close to the edge. It's all kind of confusing to me."

Richard Beasley faced previous charges in Ohio, including aggravated menacing, tampering with evidence, possession of criminal tools and illegal cultivation of marijuana, court records show. He also was convicted in Texas of burglary and unauthorized use of a motor vehicle in 1985, sentenced to a 40-year prison sentence and placed on parole for 34 years in 1989.

Beasley was in the Summit County jail Friday on drug trafficking, compelling prostitution and fugitive-from-justice charges when police made arrests in the robbery scheme. The Associated Press generally does not report the names of minors charged with crimes. A message left with an attorney for Beasley listed on court documents was not returned.

A U-Haul truck loaded with beer kegs plowed through a parking lot filled with tailgaters Saturday before the Yale-Harvard football game, killing one woman and injuring two others, authorities said.

Police spokesman David Hartman said the truck entered a parking lot designated for pre-game tailgating, sped up for unknown reasons and hit the women. The truck then crashed into other U-Haul vans in the lot.

A 30-year-old Massachusetts woman was taken to Yale-New Haven hospital, where she was pronounced dead. Another woman was seriously hurt, but was in stable condition. The third woman suffered minor injuries.

The New Haven Register reported that the driver of the U-Haul was taken into custody.

Police said there were several conflicting accounts from witnesses. Some told police it appeared the truck suddenly lurched forward; others said the driver pressed the pedal in road rage because the women weren't moving fast enough, according to the Register.
